Acetone Bio-Sniffer (Gas-Phase Biosensor) for Monitoring of Human Volatile Using Enzymatic Reaction of Secondary Alcohol Dehydrogenase

We developed a highly sensitive acetone bio-sniffer (gas-phase biosensor) based on an enzyme reductive reaction to monitor breath concentration. The device was constructed by attaching flow-cell with nicotinamide adenine dinucleotide (NADH)-dependent secondary alcohol dehydrogenase (S-ADH) immobilized membrane onto fiber-optic NADH measurement system. This system utilizes ultraviolet light emitting diode as excitation source. Acetone vapor measured the fluorescence of consumption enzymatic S-ADH. A phosphate buffer that contained oxidized circulated into rinse products and excessive substrates from optode; thus, enables real-time monitoring photomultiplier tube detects change in emitted NADH. relationship between intensity concentration identified be 20 ppb 5300 ppb. encompasses range found healthy people those suffering disorders carbohydrate metabolism.
